Hepcidin messenger RNA expression in human lymphocytes

Hepcidin之前主要的研究為其在iron metabolism的耜份

在這一篇immunology 2010的文章，研究human lymphocytes activation時，觀察到hepicdin mRNA的上升

顯示hepcidin可能在lymphocytic immunology中有其重要性

目前人體上N-acetyl L-aspartate這個acetyl amino acid在正常人體上只被在腦中發現有它的存在

這個team發現在ovarian tumours裡，在cysts fluid中藉由GC/MS確認它們的存在.
顯示出ovarian tumor會表現NAA

據作者，目前也有相當多的研究者在嘗試了解到底NAA在人體上的作用為何(or brain specifically，因為過去只在大腦中發現)

目前初步認為NAA跟大腦中水的homeostasis有關係

講到了，科學家把合成morphine的前驅物THP打進老鼠，結果就可以找到下一個中間產物，把這個東西再打進老鼠，又可以找到下一個中間產物，如此如此一直到最後在尿液中找到morphines類的trace
然後在很多tissue中也找得到morphine
顯示出有pathway可以合成morphine

->未來可能養一堆老鼠讓他們做很多的morphine??
XDDD

作者表示接下來要identify 這個pathway裡的enzymes
並且，這個process是natural就發生的，那是不是老鼠本身就會合成一些morphines, 這些morphines在那些組織上的作用到底是什麼

還蠻酷的.

ABI推出Attune acoustic focusing cytometer

傳統的cytometry現在已經很強大，利用hydrodynamic focusing來達到單一細胞通過的能力~~

這個新的cytometer不用hydrodynamic focusing.
而是使用超音波做focusing~~

超威的啊~~

我是第一次看到

主要的內容還沒有看完
但是看起來最大的優點是，功能完備 但是機器很小，差不多是微波爐大小

最近日劇真的看很少..
有在抓就是了…但是沒有特別在看..

我還是喜歡看歐美的..
主要都是靠eztv和tv.torrent.ro這個private tracker上載..

因為畫質的關係，都抓720p的..
一個40分鐘左右的一集，就要1GB

想當然的我的硬碟爆了~~XDDD
我是只要能抓我就會抓~所以就變成這種局面了..
現在外接的就有1.5TB*3

然後正在存的有一顆2TB

以後應該會繼續買2TB的
購買速度應該差不多是2個月..
真是個大開銷啊~~

因為要弄這個真的要把Apache、php、SQL等弄懂.
而且一些相關的規範，實際要做的功能、copyright等.
都還要弄懂

所以主要是會等我的事情告一段落之後，架起一個joomla site，在上面做一點交流、討論，為wiki站做準備.

未來這個wiki站應該會有一些附屬的部份
1. video server for upload of experiment tutorials
2. forum for discussion between professionals and for questions from the public
3. blog for release of news

nature news上寫最近幾篇文章address在以uv 260nm/280nm測定DNA濃度可以受到塑膠化學藥劑影響的文章

對學生物的人來說都知道有幾個uv的波段可以用來測定DNA、RNA的濃度.
主要是因為base在260nm有較大的吸收波長
而DNA、RNA品質可以藉由evaluate OD 260nm和其它波長的ratio
包括OD230/260, OD 260/280

尤其現在有nanodrop的儀器出現..只要2uL甚至1uL就可以evaluate sample中的nucleic acid濃度..

用spectrophotometry是很方便的方式，但是現在這篇文章在討論，目前在生醫實驗室中長用的塑膠容器，溶出的化學藥劑可能會造成DNA測定的誤差，甚至到300%

First author和corresponding author是University of Münster, Germany的研究者~不過作者群有很多其它研究機構的，包括Albert Einstein college of medicine、University of Glasgow, Max-Plank-institute of molecular biomedicine, Harvard等。

In a previously developed inducible transgenic mouse model of chronic myeloid leukemia, we now demonstrate that the disease is transplantable using BCR-ABL+ Lin–Sca-1+c-kit+ (LSK) cells. Interestingly, the phenotype is more severe when unfractionated bone marrow cells are transplanted, yet neither progenitor cells (Lin–Sca-1–c-kit+), nor mature granulocytes (CD11b+Gr-1+), nor potential stem cell niche cells (CD45–Ter119–) are able to transmit the disease or alter the phenotype. Thephenotype is largely independent of BCR-ABL priming before transplantation. However, prolonged BCR-ABL expression abrogates the potential of LSK cells to induce full-blown disease in secondary recipients and increases the fraction of multipotent progenitor cells at the expense of long-term hematopoietic stem cells (LT-HSCs) in the bone marrow. BCR-ABL alters the expression of genes involved in proliferation, survival, and hematopoietic development, probably contributing to the reduced LT-HSC frequency within BCR-ABL+ LSK cells. Reversion of BCR-ABL, or treatment with imatinib, eradicates mature cells, whereas leukemic stem cells persist, giving rise to relapsed chronic myeloid leukemia on reinduction of BCR-ABL, or imatinib withdrawal. Our results suggest that BCR-ABL induces differentiation of LT-HSCs and decreases their self-renewal capacity.

因為我的英文不好啦..
最後就是說BCR-ABL這個fusion gene，aka philadelphia chromosome, 是Chronic myelogenous leukemia(CML)中最常見的染色體異常，主因為t(9;22)轉位，造成的BCR-ABL fusion.

這篇主要是 在講BCR-ABL可以造成long term hematopoietic stem cell的分化，並降低該血液幹細胞的自我更新能力。

well sorry, i have been really lazy.

Here’s some new thoughts that have been on my mind for a while.

I’m thinking about creating a wiki site of clinical laboratory science.
Well someone may say.. we already have the wikipedia, why the hell would i need this new wiki site.
I have a couple of reasons.

wikipedia is really great, and i’m thinking about information tunneling between the new site and the wikipedia.
but like all other medical sites, information on the internet regarding to medicine should get some review before it can be published.
Having a logon required wiki site can provide a place for us to organize clinical laboratory science information with peer review before giving it to the public.

All patients have the right to understand what’s going on to their body.
And health care providers have been in a dilemma, patients have the right to know, but everything is possible in any medical situation, and not all of them would happen to a patient, but possible.
Should the patient understand all the possible medical outcome? Most of the time, even we, as healthcare providers, in face of a medical situation which is not in our expertise, are having problems understanding, let alone patients who have no science training before.
Giving too much information may not necessary in a patient’s best interest. BUT ethically, a patient does have the right to understand their condition.

Having a site like this may be the place to provide reliable information for those who intend to know more about their condition from a laboratory medicine approach.
This site could release articles after a series of peer review, and provide to the public to access.
The Wiki site could also make like LabTestsOnline (http://www.labtestsonline.org)
make the patient oriented approach as one of the primary objective of the site.

Also i would like to make this site as a platform for continue education for all healthcare providers(this may be too far) but at least for all clinical laboratory workers.

biomedical science have been through tremendous advances over the past few decades, and we are only going forward even faster.
With advances in molecular biology, systems biology and other principles, translational medicine is only going to thrive even faster.

Too much information is out there. And memorizing all the information teachers think is important really isn’t helping out students.
Information from biomedical research has grown tremendously, but our students still go to a 4 year college life.

How would you expect students to stuff in information much much more than before and work perfectly?
So of course there have been some problems in clinical laboratory science education in my perspective.

And at this age, i think memorizing isn’t really the important thing, the important thing should be information gathering and sorting quickly when it is presented.

The site can be a consistenly updated wiki for information with sources from books, journal papers. with appropriate citing, can be useful for all laboratory personnel to pick up easily, and refer to the paper quickly if necessary.

There are also some other features i have in mind, and i think is practical, yet a little out of the league since the site hasn’t yet existed.

so…that’s it~
